Die NucleoBond RS Säule ist eine Anionenaustausch-Säule, die für den halbautomatischen Einsatz in einer Umgebung mit hohen regulatorischen Anforderungen konzipiert ist. Die geschlossene Bauweise der Säule in Kombination mit der Verarbeitung mittels einer Peristaltikpumpe oder eines FPLC-Systems gewährleistet reproduzierbare Ergebnisse bei gleichzeitiger Minimierung des Arbeitsaufwands. Diese Säulen sind in der Lage, hochreine, hochintegrierte Plasmid-DNA in Impfstoffqualität zu isolieren, was zu unserem bisher niedrigsten Endotoxinprodukt führt (≤ 0,01 EU/µg). Je nach Säulengröße und Probenzufuhr können Ausbeuten von 10-200 mg erzielt werden. Um die Vielseitigkeit der Säule zu ergänzen, steht eine Reihe von Clean-up-Protokollen zur Verfügung, die als Support-Protokolle verwendet werden können.

Plasmids isolated with NucleoBond RS columns show very high structural integrity
The supercoiled form of plasmid DNA is considered to be the desirable form for applications like gene therapy or vaccination. Regulatory standards specify that plasmids have to be present with more than 90 % supercoiled form. These data show that plasmids isolated with our NucleoBond RS columns meet these standards.

Technical data from plasmid isolations using our NucleoBond RS columns show endotoxin levels ≤ 0.01 EU/μg DNA, A260/A280 purity ratios between 1.8 – 2.0, and percentages of supercoiled (ccc) form ≥ 90 %.
