                 High-volume sample processing on automated SPE systems 
 Solid-phase extraction (SPE) is an established method for enriching trace analytes, which can be very time-consuming. MULTI 96 plates optimize SPE workflows by reducing labor and delivering consistently high-quality data, whether used manually or in fully automated setups. However, in automated processes, sample sizes quickly reach their limits due to limited space. 
 CHROMABOND C18 100 mg MULTI 96 Monoblock SW is the first plate in the new SquareWell (SW) format in the MACHEREY-NAGEL portfolio. The modified design features 96 square instead of round cavities, increasing the sample volume to up to 2,000 µL per cavity while maintaining the same height. The plate can therefore be used without any problems in existing 96-well SPE systems.

                 Robotic sample preparation lines as well as some autosamplers, like the CTC GC / Combi PAL and comparable instruments, are constructed that way that they lift and transport vials with a magnet the size of the vial’s cap. For such instruments we have now launched a magnetic, fluorine-free 9 mm screw closure for PFAS analysis. Instead of a PTFE lamination the silicone liner is covered with polypropylene as the chemically inert barrier towards the sample, in order to guarantee sample integrity in this special application.

                 As a replacement for the discontinued 11 mm crimp closures with MN REF 702001, MACHEREY-NAGEL has added a range of new natural rubber/ETFE closures in silver (MN REF  70246 ), green ( 70246.1 ), red ( 70246.2 ) and blue ( 70246.3 ) caps to their delivery program. 
 The high restoring forces of the natural rubber allow multiple injections without sample loss. In comparison to the discontinued NR/ETFE closure 702001,  70246  has a lower durometer (45° shore A instead of 60° shore A) and thus is easier to penetrate. This offers more safety for the needle and less fragmentation during piercing (lower risk of contamination).  The ETFE lamination has a comparable chemical resistance to other types of PTFE. The main area of usage for these closures is in GC applications where instrument manufacturers like Agilent offer them as a cost-effective closure for routine analysis (see 5181-1210/100; 5183-4498/1000). The self-manufactured closure is available ex stock at sensational pricing. For those who prefer screwing instead of crimping, a screw closure N 9 with a blue open top cap and with natural rubber/ETFE liner can be obtained with MN REF  702460 .

                 Single  micro-inserts  that can be used in different types of vials are often used in analysis for such small sample quantities. Combining vials and individual inserts as separate components is cost effective, however, filling of such small containers is unhandy and fraught with risk. 
 Firmly integrated or fused-in micro-inserts in  clear  and  amber glass vials  have the advantage of a ready-to-use unit that can easily and safely be filled due to the firm fixation of the insert. Afterwards the vial can directly be placed on the lab bench or in a tray/rack. Sample removal is carried out without any risk for the needle due to the reliable centering of the micro-insert. The risk of any contamination is always lower with ready-to-use sample containers in comparison to individual components that still need to be assembled in the lab. 
 In order to further facilitate the micro-sampling process, we now offer the clear screw neck vial N 9 ( 702007.0 ) and the clear crimp neck vial N 11 ( 702891.0 ) optionally also with a writing patch.

                 Pharmacopoeias contain generally applicable rules on the quality and testing of pharmaceuticals. There are numerous pharmacopoeias worldwide. The official pharmacopoeia for the USA is the United States Pharmacopoeia (USP) and for Europe the European Pharmacopoeia (EP). 
 The monographs describe the methods to control the purity of drugs. Often drugs and their impurities are analyzed with HPLC. The pharmacopoeias also list the permitted changes to adapt a method. For example, column length and particle size can be changed within certain limits without having to revalidate a method.

                 Vitamin D stands for a group of fat-soluble vitamins, the so-called calciferols. These include the most important forms vitamin D2 (ergocalciferol) and vitamin D3 (cholecalciferol). Vitamin D plays an essential role in regulating the calcium level in the blood and in bone formation. It is also involved in other metabolic processes and in the protein biosynthesis.  For several years, it has been suspected that there are links between vitamin D levels in the body and chronic diseases. To date, however, evidence is lacking. The majority of vitamin D (approx. 80-90%) is formed in the skin with the help of sunlight (UV-B radiation); approx. 10-20% is contributed by diet, e.g. via fatty sea fish, eggs, edible mushrooms.&amp;nbsp;Therefore, especially in the winter months with little sunlight, there is a risk of undersupply and resulting low vitamin D levels. Therefore, the vitamin D value is also a common parameter in clinical diagnostics. Vitamin D level is determined by measuring 25-hydroxyvitamin D, or 25(OH)D, in blood serum. 25(OH)D is a precursor of active vitamin D. It is often also called &quot;storage vitamin D&quot;. There are various reference values for the purpose of classifying the measured values. The Robert Koch Institute, for example, refers to the classification of the Institute of Medicine (IOM) in the USA. 
 For analysis, the combination of solid phase extraction (SPE) with subsequent HPLC-MS/MS analysis is expedient. Due to the large number of samples, automation of sample preparation is an obvious solution. For this purpose, MACHEREY-NAGEL has developed an automated sample preparation in 96-well format on an Eppendorf epMotion ®  5075vt robot. CHROMABOND ®  HLB serves as a sorbent for analyte preconcentration. High recoveries and good reproducibilities were achieved.&amp;nbsp;
