Sample pretreatment:Sample is adjusted to pH 1-7 in relation to the cationic compounds
Conditions:Column conditioning: 3 ml methanol, then 3 ml water.
(Do not let run the column dry.)

Sample aspiration: The prepared sample is passed through the column by vacuum or pressure (max. 1000 ml sample volume)

Washing 1: With 3 ml AcOH / water (5:95, v/v) impurities are removed.
Washing 2 (Elution 1): With 3 ml methanol / water (20:80, v/v) neutral and acidic compounds can be removed (eluated).

Elution 2: Compounds are eluted with 2 x 2 ml methanol/acetone (1:1, v/v) + 5% NH3 or with 2 x 2 ml methanol/acetone (1:1, v/v) + 20% AcOH

Legend:All solvent volumes are based on the amount of adsorbent (200 or 500 mg) and have to be adjusted according to the size of used cartridge. This protocol is a suggestion for standard procedures and should be optimized (solvents, volumes, adsorbent amount, ....) due to necessity of the investigated sample or compound and subsequent analysis (GC, HPLC, ...).
