Sample pretreatment:Add 1.4 ml of 0.1 mol KH2PO4 buffer (pH 6) to 0.1 ml of blood serum and
centrifuge
Conditions:Column conditioning:
2 ml methanol, then 2 ml of the 0.1 mol KH2PO4 buffer (pH 6)

Sample application:
Slowly force or aspirate the clear supernatant from the sample pretreatment
through the column

Column washing:
2 ml 0.1 mol KH2PO4 buffer (pH 6), then 1 ml 0.1 mol acetic acid, then 2 ml
methanol; afterwards dry column by centrifugation (2 min., 4000 rpm.) and under
vacuum for 10 min.

Elution:
1.5 ml dichloromethane / 2-propanol / ammonia solution (25 %) (80:20:2, v/v/v)



Further analysis: For further analysis we recommend HPLC with the column
CC 250/2 NUCLEOSIL 100-5 C18 AB (Cat.-No. 721 663.20) as described in our
application 1740 'Analysis of morphine, benzoylecgonine, cocaine, codeine from
serum'

or
GC/MS after derivatisation with perfluoropropanic acid/perfluoropropanol, i.e.
with a OPTIMA 5 MS, L: 30 m, ID: 0.25 mm, FT: 0.25 m (Cat.-No. 726 220.30)
