Movie NucleoBond® Finalizer vs Qiagen® QIAprecipitator Midi

The NucleoBond Finalizer motion picture shows experiments that have been performed according to each manufacturer’s protocol. The following kits and experimental parameters have been used:
Kits used:
MACHEREY-NAGEL: NucleoBond Finalizer, in the motion picture named “MN”
QIAGEN®: QIAprecipitator Midi, in the motion picture named “Q”
Experimental parameters:
Starting material MN: 5 ml NucleoBond® Xtra Midi eluate containing 577 µg plasmid DNA
(experimental conditions see below).
Starting material Q: 5 ml Qiagen® HiSpeed® Midi eluate containing 370 µg plasmid DNA
(experimental conditions see below).

Experimental conditions for plasmid DNA purification using NucleoBond Xtra Midi and Qiagen® HiSpeed® Midi:
Plasmid: pcDNA3.1, Bacterial strain: E.coli DH5a®, Culture medium: LB (Luria-Bertani), Cultivation conditions: Incubation over night at 37°C with constant shaking (200 rpm), OD600 of the overnight culture: 6.5, Volume of the overnight culture: 50 ml (each).

Yield, purity, and concentration of the isolated plasmid DNA were determined by photometric measurement. The experiments have been performed under laboratory conditions.
