Column conditioning: 5 ml methanol, then 5 ml water
(Do not let run the column dry !)
Sample aspiration: The prepared sample is passed through the column by vacuum or pressure
Washing: With the following washing mixtures impurities are removed:
a) 2 ml 0.1 N HCl in water
b) 2 ml methanol
under circumstances c) 2 ml 2% NH3 in water
and/or d) 2 ml 30% methanol in water / 5 % NH3 in water
Drying: With nitrogen or air
Elution: Analytes are eluted with 3 x 2 ml methanol / 5% NH3
Evaporation and reconstitution (if necessary)

The volumes are typical for columns 3 ml, 200 mg. The method can be easily adjusted to different column sizes by changing the volumes in the single steps. Depending on the application the solvent for the elution can be changed and may give improved results. |
